ACE-031

ACE-031 (also known as Ramatercept, CAS: 1621169-52-5, Myostatin Inhibitory Peptide 7) is a recombinant ActRIIB-Fc fusion protein and potent myostatin/activin ligand trap developed by Acceleron Pharma (now a subsidiary of Merck). It was designed to treat muscle-wasting disorders but was discontinued due to safety issues.

Primary Targets 

  • Myostatin (GDF-8): Major negative regulator of muscle growth

Pathway Inhibition

  1. ACE-031 binds ligands in blood/extracellular space
  2. Prevents ligands from binding native ActRIIB on muscle/bone cells
  3. Blocks SMAD2/3 signaling (the inhibitory pathway that suppresses protein synthesis and satellite cell activation)
  4. Net effect: Uninhibited muscle growth, increased bone density, reduced fat

Off-Target Binding 

  • Binds BMP-9/10 (critical for vascular integrity and endothelial function)
